Electrochemical Immunoassay for Free Prostate Specific Antigen (f-PSA) using Magnetic Beads
نویسندگان
چکیده
Prostate specific antigen (PSA) is a prominent marker for the prostate carcinoma. It is found in human blood in free (f-PSA) and complex forms. These two forms together are called total PSA (t-PSA). Estimation of both forms is essential to predict malignancy. In this study we report a unique and effective technique of electrochemical detection of fPSA using magnetic beads on a three-electrode screen-printed sensor. A magnetic bead enzyme linked immunosorbent assay (ELISA) was performed in a cuvette. Following the immunoassay, magnetic beads were recovered by a magnetic concentrator and transferred on the working electrode of the 3-electrode assembly. The amperometric response, a measure of the amount of residual enzyme activity on the beads and hence the concentration of analyte in solution, was determined by addition of enzyme substrate. The device has a detection limit of < 0.1 ng mL f-PSA and a linear range of 0 to 1 ng mL f-PSA.

What is the most appropriate test in detecting prostate cancer in patients with intermediate prostate specific antigen levels?
Abstract Background: Regarding the variety of tests present for detecting and also screening prostate cancer and also bearing in mind the advantages and disadvantages of these tests we decided to re-evaluate these tests (total prostate specific antigen and all of its modifications) in detecting prostate cancer in men with intermediate serum PSAlevels. Methods: Following a cross se...

Background and Objectives: Detection of the biomarkers is one of the effective methods for diagnosis and treatment of prostate cancer. Prostate specific antigen (PSA) is currently the best biomarker available for controlling and detecting this cancer. The purpose of the current study was to design an electrochemical aptamer-based biosensor (electrochemical aptasensor) to measure...
متن کاملProstate-specific antigen kinetics after hypofractionated stereotactic body radiotherapy for localized prostate cancer
Background: stereotactic body radiotherapy (SBRT) has emerged as an effective treatment for localized prostate cancer. However, prostate-specific antigen (PSA) kinetics after SBRT has not been well characterized. The objective of the current study is to analyze the rate of PSA decline and PSA nadir following hypofractonated SBRT in localized prostate cancer. Materials and Methods: From 2008...
متن کاملIdentification of precursor forms of free prostate-specific antigen in serum of prostate cancer patients by immunosorption and mass spectrometry.
The structural features of the free prostate-specific antigen (F-PSA) present in human blood have not been clarified up to now, and it is, therefore, not known why F-PSA is not complexed by the protease inhibitors that are present in human blood in large amounts. This lack of information is mainly attributable to the low amount of F-PSA in serum, which makes the isolation and structural charact...
متن کاملFree-to-total prostate specific antigen ratio in clinical staging of prostate cancer.
The value of the free-to-total serum prostate-specific antigen (f/t PSA) ratio was compared with that of the total prostate specific antigen (tPSA) value for the prediction of clinical stage in patients with prostate cancer. The f/t PSA ratio was obtained from the frozen sera of 56 untreated patients with histologically proven BPH and 78 patients with prostate cancer. The clinical stage was org...
